The process of melanin synthesis and distribution is called melanogenesis, a process that is based on melanocytes present among the basal cells of the epidermis. Pigments formed in melanocyte melanosomes are then stored in the basal layer of epidermal cells, as well as in dermal macro-phages, which become melanophores. From the embryological point of view, melanocytes derive from the melanoblasts of the neural crest, from where they migrate during the first months of life into the skin, eye, cochlea, bone, peripheral nervous system, heart and adipose tissue. The melanic pigments, eumelanin and pheomelanin, are the final product of complex biochemical reactions starting from the amino acid L-tyrosine. Melanin has a major role in skin homeostasis through the photoprotection it offers from the harmful effect of ultraviolet radiation. Melanin absorbs and/or reflects ultraviolet radiation but is also involved in the neutralizing process of free radicals and reactive oxygen species. Pigmentogenesis is a dependent oxygen process and is controlled by intrinsic factors (genetic and hormonal) as well as extrinsic factors (ultraviolet radiation). Melanogenesis is stimulated by stimulant melanocytic hormone, adrenocorticotropin hormone, estrogens and proges-terone. The present review aimed to provide a summary of recent data about melanogenesis physiology.

The skins of phyllomedusine frogs have long been considered as being tremendously rich sources of bioactive peptides. Previous studies of both peptides and cloning of their precursor encoding cDNAs have relied upon methanolic skin extracts or the dissected skins of recently deceased specimens and have not considered the different glands in isolation. We therefore focused our attention on the tibial gland of the Giant Monkey Frog, Phyllomedusa bicolor and constructed a cDNA library from the skin secretion that was obtained via mechanical stimulation of this macrogland. Using shotgun cloning, four precursors encoding host-defense peptides were identified: two archetypal dermaseptins, a phyllokinin and a phylloseptin that is new for this species but has been recently described from the Waxy Monkey Leaf Frog, Phyllomedusa sauvagii. Our study is the first to report defensive peptides specifically isolated from anuran tibial glands, confirming the hypothesis that these glands also contribute to chemical defense. Moreover, the discovery of novel compounds for this otherwise very well characterized species suggests that this largely neglected gland might possess a different cocktail of secretions from glands elsewhere in the same animal. We will also discuss some evolutionary implications of our findings with respect to the adaptive plasticity of secretory glands.

Serum calcium levels are regulated by the action of parathyroid hormone (PTH). Major drivers of PTH hypersecretion and parathyroid cell proliferation are the hypocalcemia and hyperphosphatemia that develop in chronic kidney disease patients with secondary hyperparathyroidism (SHPT) as a result of low calcitriol levels and decreased kidney function. Increased PTH production in response to systemic hypocalcemia is mediated by the calcium-sensing receptor (CaR). Furthermore, as SHPT progresses, reduced expression of CaRs and vitamin D receptors (VDRs) in hyperplastic parathyroid glands may limit the ability of calcium and calcitriol to regulate PTH secretion. Current treatment for SHPT includes the administration of vitamin D sterols and phosphate binders. Treatment with vitamin D is initially effective, but efficacy often wanes with further disease progression. The actions of vitamin D sterols are undermined by reduced expression of VDRs in the parathyroid gland. Furthermore, the calcemic and phosphatemic actions of vitamin D mean that it has the potential to exacerbate abnormal mineral metabolism, resulting in the formation of vascular calcifications. Our senses of touch and hearing are dependent on the conversion of external mechanical forces into electrical impulses by the opening of mechanosensitive channels in sensory cells. This remarkable feat involves the conversion of a macroscopic mechanical displacement into a subnanoscopic conformational change within the ion channel. The mechanosensitive channel NOMPC, responsible for hearing and touch in flies, is a homotetramer composed of four pore-forming transmembrane domains and four helical chains of 29 ankyrin repeats that extend 150 Å into the cytoplasm. Previous work has shown that the ankyrin chains behave as biological springs under extension and that tethering them to microtubules could be involved in the transmission of external forces to the NOMPC gate. Exercise has the potential to decrease negative side effects, lower societal medical costs, and increase quality of life. Exposure to physical exercise increased participation and led the way for a variety of pilot testing movement-based interventions with a diverse, rural, clinical mental health patient population in treatment for numerous clinical diagnoses. The original research (Reuille-Dupont, 2015) is briefly outlined before a discussion of theory used to determine and perform movement-based interventions for psychological and physical health goal treatments. Throughout the paper, psychological and exercise science theory and research are overlaid to explain the physical implications and psychological shifts of movement-based treatment. It is important to understand common terminology to engage clients and other health care practitioners in movement-based treatment for psychological and physical health. In addition, as specialists in understanding the body’s role in experience, it is the somatic psychologist’s responsibility to promote and advocate for “exercise as medicine” when possible. Included are visuals to help outline and overlap the disciplines for better understanding, increased awareness, and expanding the language somatic psychologists need to engage in multidisciplinary healthcare teams.

The effect of 5-aminoimidazole-4-carboxamide-ribonucleoside (AICAR) activation of the AMP-activated protein kinase (AMPK) on the transport of the model radiolabeled dipeptide [3H]-D-Phe-L-Gln was investigated in the human epithelial colon cancer cell line Caco-2. Uptake and transepithelial fluxes of [3H]-D-Phe-L-Gln were carried out in differentiated Caco-2 cell monolayers, and hPepT1 and glucose transporter 2 (GLUT2) protein levels were quantified by immunogold electron microscopy. AICAR treatment of Caco-2 cells significantly inhibited apical [3H]-D-Phe-L-Gln uptake, matched by a decrease in brush-border membrane hPepT1 protein but with a concomitant increase in the facilitated glucose transporter GLUT2. A restructuring of the apical brush-border membrane was seen by electron microscopy. Perforated whole-cell patch-clamp recordings obtained with nystatin are frequently used to preserve intracellular integrity. However, the perforated-patch configuration may sometimes undergo a spontaneous change into the conventional whole-cell configuration, especially when lymphocytes are investigated. The electrophysiological criteria-- previously described--for establishing the existence of the perforated whole-cell configuration have been shown to be insufficient. Thus, the dye eosin, applied to the pipette solution, was tested as a tool for discriminating between the perforated and the conventional whole-cell configurations on rat T-lymphocytes. The dye never entered the cell from the pipette during the entire measurement in the perforated whole-cell configuration. In contrast, all cells in the conventional whole-cell configuration became red immediately after membrane rupture. Eosin barely changed the currents studied. The results suggest that eosin is a dye of choice for verifying a true perforated-patch configuration.

Soccer is an intermittent team-sport, where performance is determined by a myriad of psychological, technical, tactical, and physical factors. Among the physical factors, endurance appears to play a key role into counteracting the fatigue-related reduction in running performance observed during soccer matches. One physiological determinant of endurance is movement economy, which represents the aerobic energy cost to exercise at a given submaximal velocity. While the role of movement economy has been extensively examined in endurance athletes, it has received little attention in soccer players, but may be an important factor, given the prolonged demands of match play. For this reason, the current review discusses the nature, impact, and trainability of movement economy specific to soccer players. A summary of current knowledge and limitations of movement economy in soccer is provided, with an insight into future research directions, to make this important parameter more valuable when assessing and training soccer players' running performance.

istemli kasların REM uykusundaki atoni halini ve uyku-uykusuzluğun etkilerini çok güzel açıklamıştır. Dönemindeki
bilgi birikimi ve teknolojik imkânlarla yazılan bu kitapta uyku fizyolojisine ayrıca yer verilmesi de bir başka önemli
noktadır.

To enable a relevant interpretation of otolith strontium : calcium (Sr/Ca) variations in terms of habitat shifts of eels, the Sr/Ca-salinity relationship in eel otoliths was validated. Downstream and upstream migrations of young eels were reproduced in the laboratory by transferring groups of fish every 2 months between aquaria filled with water coming from the Dordogne river (salinity = 0), the upper Gironde estuary (salinity = 5), the lower Gironde estuary (salinity = 25) and the coast (salinity = 30), which represented the salinity gradient observed in the Gironde-Garonne-Dordogne watershed. Ontogenetic changes in otolith Sr/Ca were assessed in two groups of control fish that were kept in one of either two constant salinities (fresh water or seawater). X-ray electron microprobe (wavelength dispersive spectrometry, WDS) analyses of Sr/Ca ratios in the otoliths showed that the change of aquarium was recorded as a Sr/Ca increase (downstream migration) or a Sr/Ca decrease (upstream migration). No ontogenetic effect was detected in otoliths of control fish outside glass eel marks in either group of fish. The electron microprobe (WDS) analysis of the Sr/Ca life (transected in several otoliths of eels caught in the Gironde-Garonne-Dordogne watershed) showed that some of them were migrant eels that had experienced one major habitat shift during their continental life.

Ethnobotanical approaches to the study of Mediterranean food plants offer novel ways for analyzing and preserving traditional knowledge and agrobiodiversity in the Mediterranean area. This article highlights our strategy to increase the awareness within traditional knowledge systems and encourage the continuous evolution of it, avoiding the loss of substantial parts of the local cultural and biological diversity. The strategy is part of a broader stream of thought, which does attempt to disseminate information locally in a multitude of ways, e.g. through a range of publications in rural or urban zones, to people with or without formal education, to children or the elderly. This article is a very personal account of the experience of the authors, but there is an urgent need to assess the impact of such activities on a broader level, and, also, to reassess the impact researchers have on the communities. Our clear impression in all field sites has been that the simple fact that such traditional knowledge systems are the focus of scientific investigation are an essential element of giving renewed sociocultural value to such knowledge and that activities like the ones described here are of great interest to the communities we worked in.

A chronic continuous or intermittent gastrointestinal tract dysfunction, the irritable bowel syndrome (IBS), appears to be due to dysregulation of brain-gut-microbiota communication. Furthermore, the "microbiota" greatly impacts the bi-directional brain-gut axis communication. This article describes IBS in relation to similar diseases, presents the background to osteopathy, and proposes osteopathic manipulative treatment (OMT) to manage IBS. In IBS, OMT focuses on the nervous and circulatory systems, spine, viscera, and thoracic and pelvic diaphragms in order to restore homeostatic balance, normalize autonomic activity in the intestine, promote lymphatic flow, and address somatic dysfunction. Lymphatic and venous congestion are treated by the lymphatic pump techniques and stimulation of Chapman's reflex points. A simple treatment plan designed to lessen chronic pain and inflammation in IBS is presented based on current evidence-based literature. Since food itself, food allergies, and intolerance could contribute to symptom onset or even cause IBS, this article also provides dietary modifications to consider for patients.

Background: Isolated growth hormone deficiency (IGHD) provides the ideal model to characterize GHD without interference from other pituitary deficiencies or their treatment. No study has addressed the question whether adult patients with IGHD differ in clinical presentation or in responsiveness to GH replacement from adult patients with multiple pituitary hormone deficiencies (MPHD) receiving conventional replacement therapy. Patients and methods: Data were retrieved from the outcomes research database KIMS (Pfizer international metabolic database). Patients with IGHD accounted for 9.6% (274/2868) of all GHD patients. Patients were separated according to the timing of onset. In the adult-onset (AO) group, 167 patients with IGHD were compared to 1992 patients with MPHD. In the childhood-onset (CO) group, 107 patients with IGHD were compared to 602 patients with MPHD. To assess the effect of GH replacement after one year, a longitudinal sub-analysis in the AO group was performed comparing 89 IGHD patients to 1234 MPHD patients. The same study was done in the CO group comparing 66 IGHD patients to 386 MPHD patients. Because IGHD patients were significantly younger than MPHD patients, data analysis was also performed after adjustment for gender and age. Results: In the AO group, non-functioning and secreting pituitary adenomas were the most common primary diagnoses in both IGHD and MPHD. Medical history revealed a high prevalence of hypertension and fractures in both subgroups, but also of non-insulin dependent diabetes mellitus. The prevalence of obesity was high and the waist circumference was elevated. The lipid profile was unfavourable in both IGHD and MPHD. IGF-I concentration and SDS were comparable in both subgroup. Quality of life assessed by QoL-AGHDA was equally poor in both IGHD and MPHD. GH replacement therapy induced favourable changes without distinction.

Population of loggerhead turtles nesting in the Mediterranean Sea has probably evolved from the North Atlantic (NA) population, but is geographically and genetically distinct. We aggregated previously published and new unpublished data, and took two approaches to comparing these populations: an empirical one based on statistical analyses of morphological data, and a physiological one based on a Dynamic Energy Budget (DEB) model. We then analyzed causes of faster growth and maturation, but smaller size at puberty and ultimate size of the Mediterranean (MED) loggerhead turtles relative to their NA conspecifics. The empirical analysis showed that young juvenile, sexually mature, and fully grown MED individuals were consistently smaller in terms of length and mass. The physiological approach suggested physiological adaptations of the MED population to higher salinity and scarcer food availability. In particular, these adaptations included an increase in somatic maintenance needs, and a decrease in energy investment to reach and maintain sexual maturity. Our study therefore offers a mechanistic underpinning of previously observed but unexplained life-history traits, and showcases an application of DEB theory as a tool for comparative analysis of two distinct populations of the same species.

A gene encoding a methionine-rich storage protein, SnoSP2, was cloned and its complete cDNA sequence was determined in the corn stalk borer, Sesamia nonagrioides (Lepidoptera: Noctuidae). Potentially, SnoSP2 encoded a 748-amino acid protein, with a calculated molecular weight of 87.9 kDa and an isoelectric point pI = 9.41. Signal peptide of 15 amino acids is present at the N-terminus and the protein contained conserved insect larval storage protein signature sequence patterns. The deduced amino acid sequence of SnoSP2 showed the highest identity to the methionine-rich storage protein from Spodoptera litura (77%) and other methionine-rich storage proteins. SnoSP2 belongs to the subfamily of methionine-rich storage proteins (6.8% methionine, 9.5% aromatic amino acid), according to criteria of amino acid composition and phylogenetic analysis. Expression of SnoSP2 mRNA was determined using semi-quantitative RT-PCR and real-time PCR. When larvae were treated with juvenile-hormone analog, methroprene, SnoSP2 transcripts were induced. In non-diapausing conditions, the SnoSP2 mRNA presents in the beginning of fifth instar, increased dramatically during the sixth instar, peaked in the end of sixth instar, decreased in the early pupae and were very low at the late pupae. In diapausing conditions, SnoSP2 remain abundant through the pre-diapause, persists through deep diapause and disappear in the end of post-diapause phase.

Idiopathic pulmonary fibrosis (IPF) is thought to involve inflammatory infiltration of leukocytes, lung injury induced by reactive oxygen species (ROS), in particular superoxide anion, and fibrosis (collagen deposition). No treatment has been shown to improve definitively the prognosis for IPF patients. Superoxide dismutase (SOD) catalyzes the dismutation of superoxide anion to hydrogen peroxide, which is subsequently detoxified by catalase. Lecithinized SOD (PC-SOD) has overcome clinical limitations of SOD, including low tissue affinity and low stability in plasma. In this study, we examined the effect of PC-SOD on bleomycin-induced pulmonary fibrosis. Severity of the bleomycin-induced fibrosis in mice was assessed by various methods, including determination of hydroxyproline levels in lung tissue. Solid tumours account for 90% of all cancers. Gene therapy represents a potential new modality for their treatment. Up to now, several approaches have been developed, but the most efficient ones are the viral vector based gene therapy systems. However, viral vectors suffer from several deficiencies: firstly most vectors currently in use require intratumoural injection to elicit an effect. This is far from ideal as many tumours are inaccessible and many may have already spread to other parts of the body, making them difficult to locate and inject gene therapy vectors into. Second, because of cell heterogeneity within a given cancer, the vectors do not efficiently enter and kill every cancer cell. Third, hypoxia, a prevalent characteristic feature of most solid tumours, reduces the ability of the viral vectors to function and decreases viral gene expression and production. Consequently, a proportion of the tumour is left unaffected, from which tumour regrowth occurs. Thus, cancer gene therapy has yet to realise its full potential. The facultative or obligate anaerobic bacteria have been shown to selectively colonise and regerminate in solid tumours when delivered systemically. Among them, the clostridial spores were easy to produce, stable to store and safe to use as well as having extensive oncolytic ability. However, research in animals and humans has shown that oncolysis was almost always interrupted sharply at the outer rim of the viable tumour tissue where the blood supply was sufficient. These clostridial spores, though, could serve as "Trojan horse" for cancer gene therapy. Indeed, various spores harbouring genes for cancerstatic factors, prodrug enzymes, or proteins or cytokines had endowed with additional tumour-killing capability. Furthermore, combination of these "Trojan horses" with conventional chemotherapy or radiation therapies often significantly perform better, resulting in the "cure" of solid tumours in a high percentage of animals. It is, thus, not too difficult to predict the potential outcomes for the use of clostridial spores as "Trojan horse" vectors for oncolytic therapy when compared with viral vector-mediated cancer therapy for it be replication-deficient or competent. However, to move the "Trojan horse" to a clinic, though, additional requirements need to be satisfied (i) target tumours only and not anywhere else, and (ii) be able to completely kill primary tumours as well as metastases. Current technologies are in place to achieve these goals.

Unusual regions of densely packed membranous tubules known as tubular aggregates (TAs) have been observed in skeletal muscle fibres of mammals under numerous pathological conditions but also in health. Their causality is unclear. It is neither known whether TAs are destructive and should be treated or whether they have a compensating function in an endangered muscle. In spite of many similarities, the histochemical, immunocytochemical and ultrastructural characteristics of tubular aggregates do vary. Histochemistry provided an overall characteristic of TAs as membranous inclusions with a variety of enzymatic activities. Immunocytochemical evidence revealed that tubular aggregates contain miscellaneous proteins and that derive from membranes of sarcoplasmic reticulum and mitochondria. Background: Aerobic and intense exercises with an increase in free radicals cause damages at the cellular level, heart disease, cancer, and the development of aging processes, which one of its symptoms is increased serum concentrations of liver enzymes. Objective: The purpose of this study was to investigate the concurrent effect of four weeks of aerobic training and propolis supplementation on the activity of liver enzymes, including ALT, AST, and SOD in endurance athletes. Methods: Thirty-two male athletes (age: 21±1.4 years) in track and field were randomly divided into three groups: exercise group, exercise with placebo group, and exercise with supplement group. Propolis supplementation was taken as two tablets (500 mg) twice a day and aerobic exercise was performed for 4 weeks and in 24 sessions with an intensity of 60 to 65% of heart rate. The statistical method was done using one-way ANOVA and Tukey post hoc test by SPSS v. 18 software. Findings: The results showed that there was a statistically significant difference between groups in serum levels of SOD, AST, and ALT (P<0.05). There was not a statistically significant difference between the exercise group and placebo+exercise group in serum levels of SOD, AST, and ALT (P>0.05). Conclusion: The results showed that aerobic exercise alone can increase SOD levels and propolis supplementation with aerobic exercise can reduce AST and ALT serum levels and lead to improved liver cell function.

Objecti®e: The purpose of this study was to evaluate the efficacy and safety of intravaginal misoprostol for labor induction. Methods: 110 singleton term pregnancies with or without rupture of membranes were enrolled. Fraction-Ž. ated doses of misoprostol were applied 50᎐100 g , every 6 h until a maximum of three doses or beginning of labor. Ž. Results: The average interval "S.D. from vaginal application to the beginning of active labor and to delivery were, Ž. respectively, 9.5" 5.7 h and 14.8" 9.5 h. Failed labor induction was observed in two cases 2%. Cesarean section rate was 14%. The incidence of tachysystole was 18% and hypersystole 4%, but these situations were associated with Ž. abnormal fetal heart rate pattern hyperstimulation in only 3%. No maternal side effects and neonatal adverse effects were noted. Conclusions: Intravaginal misoprostol administration with low doses is an effective and safe method for labor induction in term pregnancies, with or without rupture of membranes.

A tick vector of Thogoto (THO) virus was Shown to secrete a factor in saliva which potentiates the transmission of THO virus to uninfected ticks feeding on an apparently non-viraemic host. The effect of the saliva activated transmission (SAT) factor on the virus occurred at the site of inoculation in the skin and was apparent even when the virus was introduced 3 days after the SAT factor, The results suggest that tick saliva can play an important role in disease transmission by virtue of host modification at the site of feeding.

Parathyroid hormone-related protein (PTHrP) is the major mediator of the humoral hypercalcemia of malignancy and of malignant osteolysis associated with skeletal metastases of common epithelial cancers. PTHrP secretion is regulated by the extracellular calcium concentration ([Ca(2+)](o)) in several types of normal and malignant cells. Because the [Ca(2+)](o)-sensing receptor (CaR) is a key mediator of [Ca(2+)](o)-regulated hormone secretion [e.g., of parathyroid hormone (PTH) by parathyroid chief cells], we investigated the expression of the CaR and PTHrP in normal and neoplastic glial cells and studied the effects of [Ca(2+)](o) on PTHrP secretion. Hybridization probes produced from DNA sequences have proven to be a powerful tool in the rapid and sensitive analysis of natural microbial communities. By using function-specific probes, such as those identifying genes coding for photosynthesis, the potential a microbial community has for performing a given function may be rapidly determined. Gene probes have also been used in the idelatification and isolation of a specific catabolic genotype in less than one-fourth the time required for the conventional culture enrichment technique. Species-specific probes constructed from portions of genes coding for ribosomal RNA have been used for the rapid identification and enumeration of bacterial species in environmental samples. The use of reassociation kinetics as a measure of community diversity and complexity is also discussed. The successful application of this technique to community analysis may reduce the time required from 1 year, for conventional analysis, to 2 weeks.

The Association of American Medical Colleges has encouraged educators to investigate proper linkage of simulation experiences with medical curricula. The authors aimed to determine if student knowledge and satisfaction differ between participation in web-based and manikin simulations for learning shock physiology and treatment and to determine if a specific training sequencing had a differential effect on learning. All 40 second-year medical students participated in a randomized, counterbalanced study with two interventions: group 1 ( n = 20) participated in a web-based simulation followed by a manikin simulation and group 2 ( n = 20) participated in reverse order. Lincolnshire contains some of the most important examples of small-leaved lime woodland in Britain. The trees have been managed since the 11th century by coppicing and many of the trees studied are several hundred years old. The genetic diversity of small leaved lime (Tilia cordata) is being investigated using RAPD markers to estimate the genetic relationships between trees. The RAPD technique provides a convenient method of assessing the differences in the genetic composition of related individuals when there is no DNA sequence information available. Initial results of RAPD analysis indicate that the trees from two separate woodland areas although largely similar show two separate clusters. This suggests that RAPDs will be a suitable tool in this study and will show how the genetic variation of the trees is affected by their location not only in their own wood but also within nearby ancient woodlands. Tissue culture techniques are also being developed to enable the successful micro-propagation of T. cordata. Factors such as sterilization procedures, plant growth regulator concentrations, temperatures and day length are being manipulated to produce optimum conditions for the production of explants. Using the plant growth regulators 6-benzylaminopurine (BAP) and naphthaleneacetic acid (NAA) it has been possible to induce both root and leaf material. Using tissue culture techniques to produce new seedlings would enable the woods to be managed in a way that maintains the inherent genetic integrity of the Tilia cordata.

Steady-state oxygen diffusion in flesh of apples (Malus domestics Borkh. CVS. Braeburn and Cox's Orange Pippin), Asian pears (Pyrus serotina Rehder. CVS. Hosui and Kosui), and nectarines [Prunus persica (L.) Batsch. CVS. Red Gold and Sunglo] was studied using a nondestructive method at 20C. Fruit flesh was found to exert a significant resistance to O 2 diffusion resulting in measurable O 2 gradients between tissues immediately beneath the skin and those at the fruit center for all these fruits. The magnitude of these O 2 gradients varied between crops and cultivars and depended on the respiration rate and on effective O 2 diffusivity in fruit flesh (D e ). Values of D c varied with the cultivar and were broadly consistent with intercellular space volume. The range of D e values obtained suggested that 0 2 diffusion in fruit flesh takes place in a combination of series and parallel modes in the intercellular space and fluid/solid matrix of the flesh. The results imply that O 2 diffusivity in flesh tissues must be taken into consideration in the determination of critical external O 2 level in controlled/modified atmosphere (CA/MA) storage.

X-ray microcomputed tomography (mCT) allows nondestructive visualisation of plant root systems within their soil environment and thus offers an alternative to the commonly used destructive methodologies for the examination of plant roots and their interaction with the surrounding soil. Various methods for the recovery of root system information from X-ray computed tomography (CT) image data have been presented in the literature. Detailed, ideally quantitative, evaluation is essential, in order to determine the accuracy and limitations of the proposed methods, and to allow potential users to make informed choices among them. This, however, is a complicated task. Three-dimensional ground truth data are expensive to produce and the complexity of X-ray CT data means that manually generated ground truth may not be definitive. Similarly, artificially generated data are not entirely representative of real samples. The aims of this work are to raise awareness of the evaluation problem and to propose experimental approaches that allow the performance of root extraction methods to be assessed, ultimately improving the techniques available. To illustrate the issues, tests are conducted using both artificially generated images and real data samples.

In this study we investigated the relationship between the presence of bound protamine on mouse and human sperm DNA and the level of chromomycin A 3 (CMA 3 ) and 4Ј6-diamidino-2-phenylindole (DAPI) fluorescence. This was accomplished by performing a competition assay between salmon protamine and fluorochromes on decondensed spermatozoa that had their nuclear proteins extracted and were fixed on slides. Various concentrations (0, 0.005, 0.0225, 0.05, 0.225, 0.5 and 5 mg/ml) of salmon protamine were added to either the CMA 3 or DAPI staining solutions. Fluorescence emission measurements of stained sperm nuclei were then performed using a microfluorometer. When the treated decondensed sperm heads were stained with either CMA 3 or DAPI all spermatozoa were found to fluoresce intensely. The addition of protamines to the spermatozoa led to an elimination of CMA 3 fluorescence, while the intensity of DAPI staining was decreased to~50% at the highest concentrations of protamine. The addition of increasing amounts of salmon protamine also induced the sperm nuclei to regain their initial condensed appearance. This study shows that protamine retains a strong affinity for sperm DNA in situ and that CMA 3 fluorescence is a strong indicator of the protamination state of spermatozoa.

We investigated whether spiders fed lipid-rich rather than protein-rich prey elevate metabolism to avoid carrying excessive lipid deposits, or whether they store ingested lipids as a buffer against possible future starvation. We fed wolf spiders (Pardosa prativaga) prey of different lipid:protein compositions and measured the metabolic rate of spiders using closed respirometry during feeding and fasting. After a 16-day feeding period, spider lipid:protein composition was significantly affected by the lipid:protein composition of their prey. Feeding caused a large and fast increase in metabolism. The cost of feeding and digestion was estimated to average 21% of the ingested energy irrespective of diet. We found no
difference in basal metabolic rate between dietary treatments. During starvation ˙VO2 and ˙VCO2 decreased gradually, and the larger lipid stores in spiders fed lipid-rich prey appeared to extend survival of these spiders under starvation compared to spiders fed protein-rich prey. The results show that these spiders do not adjust metabolism in order to maintain a constant body composition when prey nutrient composition varies. Instead, lipids are stored efficiently and help to prepare the spiders for the long periods of food deprivation that may occur as a consequence of their opportunistic feeding strategy.
